摘要
目的:通过不同理化因子对黄花乌头花药愈伤组织诱导研究,为国家一类新药关附甲素的工业化生产提供新的原料来源.方法:以黄花乌头花药为外植体,考察不同植物激素及其配比、不同种类的培养基、碳源、NH+4与NO-3比例、天然添加物及低温处理对花药愈伤组织诱导的影响.结果:2,4-D2mg/L,KT0 2mg/L为最佳激素配比;MS培养基诱导愈伤组织效果最好;以蔗糖为碳源,ρ(NH+4)/ρ(NO-3)为4∶1时有利于愈伤组织的诱导;分别以0 03mmol/L的组氨酸、赖氨酸、色氨酸、谷氨酰胺代替MS培养基中的甘氨酸有利于愈伤组织的诱导,而丝氨酸对其有抑制作用;水解酪蛋白能提高愈伤组织诱导率;低温(4~10℃)处理48~120h能促进愈伤组织生成.结论:植物细胞工程技术是解决黄花乌头原料匮乏问题的可行方法之一.
Aim: To get new resource of guanfu base A through the technology of plant cell engineering. Methods: Anthers of Aconitum coreanum were used as the e xplants. The media were appendixed with 2,4-D 2 mg/L, KT 0.2 mg/L. The different factors that influenced callus induction, such as auxin and cytokinin, media, c arbon source, proportional between NH+4 and NO-3, amino acids, natural a dditives, and cooling treatment were examined. Results: the comb ination of 2,4-D 2 mg/L and KT 0.2 mg/L is the most suitable ratio of auxin and cytokinin. MS medium was the best medium for callus induction. The optimum carbo n source was sucrose and the proportional ratio of NH+4/NO-3 was 4∶1. H istidine, lysine, tryptophan, glutamine can be used instead of glycine in the me dium of MS. But inhibition effect was shown if using serine instead of glycine. The inductive rate of callus could be increased by casein hydrolysate. Treatmen t of lower temperature (4~10℃) between 48~120 h could enhance the formation o f callus. Conclusion: The technology of plant cell engineering i s one of the suitable methods to get guanfu base A.
