低能离子注入花生四烯酸产生菌诱变选育及其产业化研究 被引量:15

摘要 利用低能离子束生物技术对花生四烯酸产生菌(Mortierella alpina,N7)进行诱变选育,并对获得的高产菌株进行了产业化研究。实验表明,在离子注入(10keV,3×1014N+/cm2)条件下,后代菌株离散程度明显高于自然分离。经连续诱变处理,最终获得一株花生四烯酸高产菌I49-N18。该菌株花生四烯酸得率高达4.66g/L,比对照N7菌株产量提高126.2%。通过微生物发酵法规模化生产花生四烯酸油脂,50吨发酵罐工业化生产,花生四烯酸产量平均达到5.11g/L。 Arachidonic acid (5, 8, 11, 14- eicosatetraenoic acid, AA) is an essential dietary component for human beings. AA, widely existing in the animal kingdom, however, the yield is only 0.2% (w/w) or lower, so it is very difficult to be applied in an industrial scale. In order to increase the yield of AA, our laboratory has focused on the study of AA fermented by Mortierelle alpina. By N+ implantation(10Kev,3 × 1014N+ /cm2), I49-N18, a high-yield AA producing strain was screened out. The AA yield of the high-yield strain was 4.66g/L. The results indicated that ion implantation had remarkable mutagenic effect on microorganism. Arachidonic acid was produced in large-scale by I49-N18. The results of production in 50-ton fermentor showed that the biomass, lipid in biomass, and AA in lipids and AA yields were 2.64g/100ml, 40.5% (w/w), 47,98% (w/w) and 5. 11g/L, respectively, which indicated that the yield of AA fermented by in industrial-scale was close to, even better than, the level of AA in the shaking batch culture. Gas chromatography/ Mass spectrograph tests showed that oil contained polyunsaturated acids such as AA、γ-Linolenic acid and Linoleic acid.
