摘要
目的 探讨脑低灌流大鼠模型局部脑血流量 (rCBF)和血脑屏障超微结构的变化。 方法 结扎Wistar大鼠 (共18只 )一侧颈总动脉 (CCA) ,围手术期以激光多普勒血流测定仪 (LDF)实时监测rCBF的变化 (以LDF测定值表示 )。分别饲养3d、3周和3个月后 ,取样在电镜下观察血脑屏障的变化。结果 (1)阻断一侧CCA后 ,LDF值下降幅度为 (25.96±1.91) %(P<0.01);(2)大鼠无明显行为异常;(3)慢性期(3个月)脑缺血大鼠实验侧出现星形胶质细胞足突空泡化异常。 结论 单侧CCA阻断后同侧rCBF下降 ,处于慢性低灌流状态 ,可致血脑屏障超微结构发生病理性改变 ,主要表现为星形胶质细胞足突空泡化异常。
Objective To evaluate the alterations of regional cerebral blood flow and ultra structure of blood-brain barrier in different stages of ischemia induced by non-infarctional hypoperfusion.Methods The forebrain hypoperfusion models were performed by ligating of unilateral common carotid artery(CCA)in18Wistar rats.The regional cerebral blood flows(rCBF)were monitored perioperatively by laser-Doppler flowmeter(LDF)Pathological ultra structural changes of blood-brain barrier induced by hypoperfusion in difˉferent stages(acute:3days,sub-acute:3weeks and chronic:3months)were assessed with transmission elecˉtron microscopy.Results(1)The mean reduction amplitude of the rCBF value by LDF monitoring was(25.96±1.91)%(P<0.01)after ligation of unilateral CCA.(2)There was no significant abnormality in behaviors of all animals after CCA ligation.(3)The transmission electron microscopy showed the marked vacuolar degeneration of the end feet of astrocytes in the chronic stage of hypoperfusion.Conclusion The pathological alterations of blood-brain barrier may develop in response to chronic cerebral hypoperfusion after unilateral CCA ligation in animal models.
