摘要
水提法提取甘薯多糖的优化工艺条件为 :提取温度 85℃ ,加水比 1∶7,提取时间 2 .5h ,提取率为2 6 .71% ;甘薯多糖经乙醇沉淀、Sevage法除蛋白和SephadexG - 10 0柱层析纯化 ,得到 4种纯多糖组分 ,其分子量分别为 86KD、4 7KD、2 3KD、176D ,百分含量分别为 18.5 1%、36 .80 %、17.93%、2 5 .2 0 % ;甘薯多糖经完全酸水解后 ,用纸层析法分析其单糖组成为葡萄糖、半乳糖和木糖。
The optimal extracting conditions of sweetpotato polysaccharide by water extraction are revealed as follows: extracting temperature 85℃, ratio between sweetpotato and water 1∶7, extracting time 2.5h and extracting rate 26.71%. By means of ethanol precipitation, protein removal by Sevage method and Sephadex G-100 column chromatorgraphy, four pure polysaccharide compositions were obtained. The molecular weight is 86KD,47KD, 23KD and 176D and the percent content is 18.51%, 36.80%,17.93% and 25.20%,respectively. After thorough hydrolysis with acid and analysis with paper-chromatography, the compositions of sweetpotato polysaccharide are glucose, galactose and xylose.
