摘要
以 3个云南省大面积推广的玉米杂交种及其亲本为试材 ,从 96对SSR引物中筛选出 2 4对引物 ,这些引物分布在玉米基因组的 10条染色体上 ,每对引物可以检测到 1~ 6个数目不等的多态性片段 (allele) ,共 97个多态性片段 ,平均为 4 .0 4个 ,片段大小介于 85~ 30 0bp之间。在比较分析各试材图谱的基础上 ,探讨了构建品种指纹图谱的统计学方法 ,建立了适于种子检验中使用的标准SSR标记指纹图谱。对随机挑选的 4个自交系进行了指纹图谱构建的理论与实践验证。
Three maize hybrids and their parents applied in Yunnan were analyzed by SSR marker to establish the standard DNA fingerprinting for seed test. Twenty four pair primers out of ninety six pair primers was distributed on ten chromosomes in maize genome. One allele to six alleles were detected by each pair primers. There were ninety seven alleles detected in total. The average of the allele detected was 4.04 per pair primer. The band range in size was from 85-300 bp. According to the data above mentioned, the fingerprinting of the maize parents was established and numbered firstly. Then the paper introduced the method to calculate probability for the fingerprinting of each parent, and finally the standard, exclusive fingerprinting of maize hybrids and their parents was determined to distinguish the truth and falseness of samples in seed test. And the fingerprinting had be verified by SSR analyzing of 4 inbred lines selected randomly.
