乙肝相关性肝癌及癌旁组织p73基因表达及突变的研究

摘要 目的 探讨p73基因在人乙肝相关性肝癌组织中的表达及其意义。方法 采用RT PCR方法检测p73基因mRNA在癌组织、癌旁组织及正常肝组织中的表达差异 ;采用SSCP检测方法比较分析p73基因在肝癌发生中的变异情况。结果 癌组织中p73基因mRNA的转录水平明显高于癌旁组织及正常组织 (P <0 0 5 ) ;然而 ,肝癌组织、癌旁组织及正常肝组织中p73基因PCR扩增片段的多态性是一致的 ,电泳未见异常条带 (P >0 0 5 )。结论 新近发现的肿瘤抑制基因p73基因在乙肝相关性肝癌组织中末见明显的基因突变 ,相反表达水平明显增高 ,可见 。 Objective To determine the expression and mutation of p73 gene in human HBV associated hepatocellular carcinoma and adjacent tissues.Methods The expressions of p73 gene were observed by RT PCR among liver cancer and tissues adjacent to tumor and normal liver tissues;at the same time,mutations of p73 gene were also examined by SSCP method.Results Level of transcripts of p73 gene in liver cancer was obviously higher than that in adjacent tissued or normal liver tissues ( P <0.05);However,polymorphisms of amplificated fragments of p73 gene among these three tissues were with one accord,with no differences( P <0.05).Conclusions No obvious gene mutation of p73 gene,which was a newly discoved tumour suppressor gene,was found,oppositely level of expression of p73 gene was apparently increased,so p73 gene may involve in the development of HBV associated hepatocellular carcinoma in a high expression manner.
