牛蒡子及其提取物改善糖尿病大鼠肾脏病变作用机制的实验研究 被引量:14

摘要 目的:研究牛蒡子及其提取物对链脲佐菌素(STZ)糖尿病大鼠肾脏病变的作用并筛选其最佳提取成分。方法:以链脲佐菌素(STZ)诱导的糖尿病大鼠为动物模型,中药治疗组分别给予牛蒡子粉饲料、牛蒡子水提物、醇提物进行6W的实验,观察了大鼠的一般状况,常规生化指标,放免法测定尿微量白蛋白的变化,用免疫组化技术测定肾皮质尿激酶型纤溶酶原激活物(u-PA)及其抑制物PAI-1的表达,激活蛋白1(AP-1)表达的改变。结果:牛蒡子及其提取物能明显改善STZ大鼠多饮、多食和体重增加减少等症状;降低尿微量白蛋白;肾小球PAS染色阳性基质面积比以及PAI-1,c-fos,c-jun的表达;减轻肾脏病理损害。结论:牛蒡子及其提取物对糖尿病大鼠肾脏病变有一定的改善,其作用机制可能与降低尿微量白蛋白,减少肾脏的PAI-1,c-fos,c-jun表达等有关。且牛蒡子醇提物较其水提物和粗粉疗效更为显著。 Objective:To investigate the influence of Fructus Arctii and its extracts on the renal pathological changes of diabetic rats and explored the protective mechanism so as to choose the most effective component. Metheds:Estabhshing the animal diabetic experimental model in Wistar rats with streptozotocin(STZ), the extract and the powder of the Fructus Arctii were given to the diabetic rats for six weeks.the changes of general condition, routine biochemical indexes, urine microalbmin, and the body weight were observed. The expressions of immunohistochemical staining was used to observe the expression of u-PA and PAI-1,c-jun and c-fos. Results:Rats of the model group had the symptoms such as polydipsia,polyphagia and the reduction of body weight gain.The extract and the powder of the Fructus Arctii could meliorate polydipsia, palyphagia, decreased urine microalbmin; lowe glomerular PAS stain positive matrix area rate and the expression of PAI-1、c-jun and c-fos;elevate the expression of u-PA in DN rat's kidneyt; relieve the renal pathological lesion,and the extract of the ethanol had better effect than its concentrated and powder. Conclusion:The ethanol extract of the Fructus Arctii relieved renal pathological changes of diabetic rats,and the extract of the ethanol of the Fructus Arctii was more effective than its concentrate and powder. The mechanism perhaps was related to all these changes.
