川芎嗪对瘢痕成纤维细胞形态和增殖抑制的影响 被引量:17

摘要 目的 探索川芎嗪治疗增生性瘢痕的机理。方法 将含川芎嗪的培养液加入增生性瘢痕成纤维细胞中 ,培养增生性瘢痕成纤维细胞 2 4h ,与未加药者对照比较。结果  1)含川芎嗪组成纤维细胞形态发生变化 ,由长梭形变为圆形 ,而培养上清中的乳酸脱氢酶 (LDH)活性及MTT(四甲基偶氮唑 )比色法结果 ,用药组与对照组均无显著差异 (P >0 .0 5 ) ;2 )流式细胞仪检测各时相细胞DNA水平 :用药组G0 +G1期细胞的百分比明显高于对照组 (P <0 .0 1) ,而G2 +M期百分比则显著低于对照组(P <0 .0 1)。结论 川芎嗪能改变增生性瘢痕成纤维细胞形态和抑制其增殖 。 Objective To explore the possible mechanism of Tetramethyl pyrazine when treating t hypertophic scar (HS).Methods Put the cultured HS fibroblasts derived from patients into the culture-flask containing SM for 24 hours, and then compare it with the control group without SM. Results We discovered that (1)the spindle-shape cells became retraction and round. Lactate dehydrogenase (LDH) activity and MTT assays showed no significant difference between the two groups (P>0.05). (2)The percentage of the DNA level in the G 0+G 1 phase, analyzed by the means of flowing cytometry, was higher than in the control group (P<0.01), while in the G 2+M phase the percentage in the former group was lower than the control group(P<0.01).Conclusions The SM can cause the alteration in cell morphon of HS fibroblast without damaging its viability and exert an inhibitory effect on fibroblast proliferation.
