摘要
丽格秋海棠组织培养试验结果表明,利用丽格秋海棠叶片作外植体是较好的取材部位;愈伤组织诱导分化的较佳初始培养基为MS+1mg/L6-BA+0.2mg/L2,4-D+3%白糖+0.4%琼脂或MS+1mg/L6-BA+0.1mg/LNAA+3%白糖+0.4%琼脂;适宜继代增殖培养基为MS+1mg/L6-BA+0.02mg/LNAA+3%白糖+0.4%琼脂;适宜生根培养基为1/2MS+1mg/LNAA+3%白糖+0.4%琼脂;适宜移栽的基质配方为河沙∶珍珠岩=2∶1。移栽适宜温度为18~20℃。
The experimental results on tissue culture of Begonia clatior showed that using the leaves as explants was best. The appropriate medium for inducing callus from explants is MS with 1mg/L 6BA + 0.2mg/L 2,4D, 3% sucrose and 0.4% agar, or MS with 1mg/L 6BA + 0.1mg/L NAA+3% sucrose +0.4% agar. The appropriate medium for adventitious buds multiplication is MS with 1mg/L 6BA+0.02mg/L NAA, 3% sucrose and 0.4% agar. The best medium for rooting is 1/2MS with 1mg/L NAA, 3% sucrose and 0.4% agar. The best component ratio of medium for transplanting plants is sand(2) ∶ perlite(1). The best temperature for transplanting is 18~20℃.
