摘要
目的 :观察及评价复合益生菌制剂“海生元”对实验性肠菌群失调症的纠正作用。方法 :1)制备复合益生菌制剂“海生元” ;2 )制备SPF·BALB/c小鼠肠菌群失调症模型 ;3)观察“海生元”对模型小鼠 (复方制剂 )组灌胃治疗效果 ;设自然恢复对照组和生理盐水对照组 (安慰组 ) ;动态比较疗后第 2、4、6天各组腹泻症状恢复情况和肠菌群定量分析。结果 :1)小鼠肠菌群失调症造模效果分析 :氨苄青霉素造模前 ,10 0 %小鼠为正常大便 (无腹泻 ) ;造模第 3天 ,6 0 %为重度 (Ⅲ度 )腹泻 ,4 0 %为中度 (Ⅱ度 ) ,即 10 0 %模型小鼠出现中、重度腹泻 ,表明造模成功。此外 ,模型小鼠肠菌群中双歧杆菌、乳杆菌未检出 ,肠杆、肠球、类杆菌数量大大低于正常小鼠 (P <0 0 1)证实出现了菌群失调症。 2 )各组腹泻恢复情况 :疗前各组均为中、重度腹泻。疗后第 2天 ,复方制剂组Ⅱ~Ⅲ度腹泻降至 4 7% ,而自然恢复组和生理盐水组分别为 99%和93% ;疗后第 4天 ,复方制剂组Ⅱ~Ⅲ度仅 16 % ,两个对照组则分别为 6 5 %和 5 7% ;疗后第 6天 ,复方制剂组完全正常者为 89% ,而两对照组分别为 2 1%。 3)各组肠菌群动态定量分析 :自然恢复和生理盐水两个对照组 ,造模后第 2、4、6天五种肠道正常生理菌数量逐渐递升 。
Objective:To observe and evaluate the correcting action of a complex-probiotic-preparation(Hai Sheng Yuan)on experimental intestinal flora imbalance. Methods:After the mice model of experimental flora imbalance were made by ampicillin,they were divided into preparation-taken group(treated by saline gastric perfusion),and natural recovery group(treated by nothing). The dynamic comparsion of the diarrhea symptom and the intestinal flora quantitative analysis were preformed among the groups at 2?4?6 day after treating.Results:(1)Analysis of model making result:After treating by the ampicillin gastric perfusion for 3 days,all of the model mice had the moderate (Ⅱ°)and severe(Ⅲ°)diarrhea(100%)and the Bifidobecterium and Lactobacillus could not be detected while Enterobacterium,Enterococcus and Bacteroid were lower noticeably than that of the normal mice(P<0.01). The result indicated that the experimental intestinal flora imbalance's model making was successful. (2)The state of diarrhea recovery:At the second day after treating,the rate of Ⅱ°~Ⅲ° diarrhea of the preparation taken group decreased from 100% to 47%,but the rate of the saline taken group and natural recovery group,93%and 99%respectively. At the forth day after treating,the rate of Ⅱ°~Ⅲ° diarrhea of the preparation-taken group was only 16%,while the rate of the saline-taken group and natural recovery group,65%and 57%respectively. At the sixth day after treating,the rate of normal feces of the preparation-taken group was 89%while the two control groups were 21%and 21% respectively.(3)The dynamic comparison of the intestinal flora quantitative analysis:At 2?4?6 day after treating,the number of the five physiological microflora Enterobacterium,Enterococcus,Bifidobecterium,Lactobacillus and Bacteroid of the two control groups increased step by step,but were still obviously lower than that of normal mice. At the second and forth day after treating,the enterobacteria and bacteroid of the preparation-taken group were higher(P<0.05) while Bifidobecteria and Lactobacillus much higher than that of the two control groups(P<0.01). As the sixth day after treating,the Enterobacteria was higher (P<0.05) while the four others were much higher than that of the two controls groups(P<0.01). Conclusion:As a microecological modulator(Synbiotics),'Hai Sheng Yuan' has the rapid and definite correcting action on treating the experimental intestinal flora imbalance. But the detailed mechanism of the preparation's therapeutieal effect should be explored further.
