摘要 对高效液相色谱(HPLC)法定性定量测定发酵液中丙酮酸含量进行了研究,确定了最佳操作条件:SCR 101H色谱柱、柱温20℃、高氯酸水溶液为流动相、流速0.8mL/min、pH3.8。通过测定回收率、重现性等项指标及与酶法测定结果的比较,证明此方法对发酵液中丙酮酸的检测具有准确、快速等特点,适于发酵液中丙酮酸的检测。 This paper studied the method which determined the contents of pyruvate in broth by HPLC. The optimal chromatographic conditions was established, which consisted of chromatographic column SCR101H, temperature 20 ℃, flow phage perchloric acid liquor, flow rate 0.8 ml/min, pH3.8. It was proved that HPLC was fit for the determination of pyruvate in broth because of its accuracy and speediness by the tests of reproductivity and recovery rate.
