摘要
目的 探讨胃癌组织中Grp94的表达及其与胃癌病理类型、临床分期及分化程度等的关系。方法 胃癌切除手术标本 3 0例 ,用半定量逆转录 聚合酶链反应 (RT PCR )检测Grp94mRNA相对表达量 ;免疫组织化学方法检测Grp94蛋白表达情况。结果 半定量RT PCR显示癌组织与癌旁组织中都有Grp94mRNA表达 ,癌组织为 0 .5 2± 0 .12 ,癌旁组织为 0 .14± 0 .0 4。前者的表达水平显著高于后者 ,其表达程度与肿瘤大小成正比 ,并随着细胞分化的降低而增高 ,Ⅲ、Ⅳ期病人显著高于Ⅰ、Ⅱ期 ;与患者性别、年龄、转移无关。免疫组织化学结果为 :癌组织大多数表达在 ～ ,癌旁组织大多表达在 -～ ,癌组织表达程度显著高于癌旁正常组织。Grp94蛋白表达程度与肿瘤大小、细胞分化、临床分期有关。结论 胃癌组织的Grp94表达呈增高趋势 ;应用Grp抑制剂或应用反义抑制战略可能抑制中晚期胃癌的生长 。
Objective To study the expression of Grp94 in human gastric cancer specimens and its clinical significance.Methods Thirty cases of human gastric cancer specimens were collected from the patients who received operation for gastric cancer from May 2001 to Nov.2002.Semi-quantitative PT-PCR assay and immunohistochemistry were used to detect Grp94 mRNA and Grp94 protein expression respectively.Results Grp94 mRNA and Grp94 proteins were overexpressed in gastric cancer specimens.The expression levels of Grp94 in cancer and normal tissues were 0.52±0.12 and 0.14±0.04 respectively (P<0.001).Immunohistochemistry revealed that the level of Grp94 protein expression in cancer was higher than in normal tissue.The levels of Grp94 expression were correlated with size,clinical stage and malignancy of caner,but not with patient's sex,age and situation of metastasis.Conclusion The levels of Grp94 are elevated in human gastric cancer specimens; Down-regulation of Grp94 expression may inhibit progress of later stage of gastric cancer and may increase the sensitivity of gastric cancer to the chemotherapy.
