摘要
目的 :观察海嘧啶对小白鼠S180肿瘤和EAC肿瘤细胞P5 3蛋白表达的影响。方法 :将含有传代后的小鼠S180肿瘤细胞和EAC肿瘤细胞的培养瓶各分为A ,B1,B2 ,B3 ,B45组 ,B1~B4组分别加入海嘧啶 ,使培养液中药物的终浓度分别为 5 0mg/L、10 0mg/L、2 0 0mg/L、4 0 0mg/L ,A组加入等体积的培养液 (对照组 )。用Westernblot法测定瘤细胞P5 3蛋白表达。结果 :S180肿瘤细胞和EAC肿瘤细胞P5 3蛋白表达率分别是 10 0 %和 90 % ;海嘧啶 10 0~4 0 0mg/L时能抑制肿瘤细胞P5 3蛋白的表达 ,与对照组比较 ,差异有统计学意义 (P <0 .0 5 )。结论 :海嘧啶抗肿瘤的药理机制与其能够抑制肿瘤细胞突变型P5 3蛋白表达有关。
Aim: To investigate the influence of sea pyrimidine on the expression of P53 protein of mouse S180 and EAC tumor cells.Methods:S180 tumor cells and EAC tumor cells were allocated into 5 groups (groups A, B 1,B 2, B 3,and B 4), respectively and sea pyrimidine with different doses were added to groups B 1 to B 4, the expression of P53 protein was detected by Western blot.Results: The positive expression rate of P53 protein were 100% in S180 tumor cells and 90% in EAC tumor cells, and sea pyrimidine with effective concentration inhibited the expression of P53 protein of mouse tumor cells.Conclusion: The anti tumor pharmacologic mechanism of sea pyrimidine is related to its inhibiting the expression of P53 protein of mouse tumor cells.
