摘要
胃癌是消化道最常见的恶性肿瘤之一,其发生是多因素、多基因相互作用的结果。随着近年来对人类基因组研究的不断深入,DNA错配修复基因缺陷(defective mismatch repair gene,dMMR)引起的微卫星不稳定性(microsatellite instability,MSI)在胃癌发生中的作用机制也越来越受到关注。错配修复(mismatch repair,MMR)基因能识别并纠正DNA在复制和转录过程中出现的错误,若其突变或缺陷使遗传错误不能被及时纠正,将引起MSI和癌基因的突变。因此,MSI和MMR有可能成为未来研究胃癌发病机制和靶向治疗的新方向。
Gastric carcinoma is one of the most common malignant tumors of the digestive system,and its occurrence is the result of multi-factor,multi-gene interactions.With the deepening research on human genome in recent years,it has been found that microsatellite instability(MSI)caused by defective mismatch repair gene(dMMR)is closely related to gastric cancer.Mismatch repair(MMR)gene can identify and correct DNA mistake made during in the process of replication and transcription,and if its mutation or defect causing genetic errors cannot be corrected in time,manifesting in MSI and cancer-related gene mutations would occur.The role and mechanism of MSI and MMR are expected to become a new direction for the pathogenesis and target treatment of gastric cancer.
