摘要
目的探讨糖足方对糖尿病大鼠足部肌腱组织转化生长因子β1(TGF-β1)、髓过氧化物酶(MPO)的影响。方法将Wistar大鼠随机分为正常组、模型组、中药预防组、中药治疗组、西药治疗组,并采取腹腔一次性注射链脲佐菌素(STZ)复制Wistar大鼠糖尿病模型。中药预防组在成模后即灌胃糖足方煎剂,中药治疗组、西药治疗组(予氨基胍水溶液)均在成模后1个月灌胃给药,至第12周。于实验第12周处死,观察大鼠后爪部肌腱组织病理变化及肌腱组织TGF-β1、MPO水平。结果中药预防组和中药治疗组均可明显改善糖尿病大鼠后爪部肌腱组织病理损害,减少组织炎症细胞浸润,且预防组优于治疗组,而西药无此作用;中药预防组、中药治疗组和西药组均可降低MPO,而以中药预防组最为显著;中药预防组和中药治疗组均可显著降低TGF-β1,而西药治疗组无此作用。结论糖足方对糖尿病大鼠足部肌腱组织微炎症反应有保护作用。
Objective To investigate the influence of Tangzu Formula on transforming growth factor-1( TGF-β1) and myeloperoxidase( MPO) in tendon tissues of diabetic rats. Methods Wistar rats were randomly divided into the normal group,model group,Chinese medicine prevention group,Chinese medicine treatment group and western medicine treatment group. The diabetic rat model was established by single intraperitoneal injection of streptozotocin( STZ). After modeling,the Chinese medicine prevention group was given the Tangzu Fomula Decoction by intragastric administration immediately,while in Chinese medicine treatment group and western medicine treatment group( treated with amino guanidine solution),the rats were treated with the corresponding drugs by intragastric administration one month after modeling. The rats were sacrificed at the 12 th week,and the pathological changes of posterior claw tendon,levels of TGF-β1 and MPO in tendon tissues were observed. Results Chinese medicine prevention group and Chinese medicine treatment group obviously ameliorated the pathological injury of posterior claw tendon in diabetic rats and decreased the inflammatory cells infiltration,and the effects of Chinese medicine prevention group were better than those of Chinese medicine treatment group,while western medicine showed no such effects. Chinese medicine prevention group,Chinese medicine treatment group and western medicine treatment group could obviously decrease the MPO level,and the Chinese medicine prevention group showed the most significant effect. The Chinese medicine prevention group and Chinese medicine treatment group could obviously decrease the level of TGF-β1,while western medicine showed no such effect.Conclusion Tangzu Formula shows protective effects on the microinflammation reaction in the foot tendon tissues of diabetic rats.
