GABA_A受体α亚单位在糖尿病性神经病理性痛大鼠背根神经节中的表达 被引量:3

The expression of GABA_A receptor α subunit in dorsal root ganglia of diabetic neuropathic rat

摘要 目的:研究γ-氨基丁酸A型受体(GABAA)在糖尿病致神经病理性疼痛(PDN)大鼠模型背根神经节(DRG)中的表达和功能。方法:正常健康雌性大鼠随机分为对照组(Control)和链脲佐菌素干预组(STZ),利用腹腔注射STZ的方法制备PDN模型,采集尾静脉血测量大鼠的血糖含量;应用热板实验检测热刺激缩足反射潜伏期;应用全细胞膜片钳技术检测PDN大鼠的L4-L6DRG神经元GABA受体介导的电流变化;应用Western Blot技术检测模型GABAA受体α亚基表达情况。结果:与对照组相比,PDN的血糖浓度上调(P <0. 05);热刺激缩足反射潜伏期缩短(P <0. 05);DRG神经元GABA受体介导的内向电流明显减小(P <0. 01);DRG神经元GABAA受体α亚基的表达量明显下调(P <0. 01),磷酸化的GABAA受体α亚基表达量明显上调(P <0. 01)。结论:糖尿病致神经病理性疼痛时,GABAA受体α亚基发生磷酸化,GABA介导的突触前抑制作用减弱。 Objective:To investigate the function and expression of dorsal root ganglia GABA_A receptorαsubunit in painful diabetic neuropathy(PDN)rat.Methods:Normal healthy female rats were divided into control group and streptozotocin intervention group randomly.PDN rat model was prepared by intraperitoneal injection of streptozotocin.The change of blood glucose was detected by model rat veins of the tails.The whole-cell patch clamp recording technique was used to clarify the change of GABA-mediated currents in DRG.Western Blot technique was used to study the expression of GABA_A receptorαsubunit.Results:Compared with control,diabetic neuropathy pain rat mode blood glucose were enhanced(P<0.05),TWL were abbreviated(P<0.05).GABA mediated inward currents were reduced obviously(P<0.01),the expression of GABA_A receptorαsubunit was down-regulated(P<0.01),but phosphorylation ofαsubunit protein was up-regulated(P<0.01).Conclusion:Because of GABA_A receptorαsubunit phosphorylation during PDN,GABA presynaptic inhibition was reduced.
