摘要
目的观察脑梗死大鼠急性期胃黏膜变化,研究通窍活血汤对脑梗死急性期胃组织的保护性作用。方法采用线栓法制备大脑中动脉闭塞局灶性脑缺血大鼠模型,将所有大鼠分为正常对照组、假手术组、脑梗死组、西咪替丁组、通窍活血汤常规剂量组、通窍活血汤低剂量组,各组分别给予相应的药物灌胃。每日1次,连续灌胃4 d后处死大鼠,分离胃组织进行电镜观察并检测胃泌素(GAS)变化情况。结果脑梗死组GAS水平较其余各组均明显偏高(P<0.01);通窍活血汤常规剂量组GAS水平比低剂量组偏低(P<0.01),比西咪替丁组降低更明显(P<0.01)。与正常对照组及假手术组相比,脑梗死大鼠的胃黏膜结构出现明显的组织破坏,通窍活血汤常规剂量组、低剂量组与西咪替丁组均能减轻上述组织破坏,且通窍活血汤常规剂量组的疗效优于西咪替丁组。结论脑梗死急性期胃黏膜出现明显损害,且与GAS增高密切相关;通窍活血汤对急性脑梗死导致的胃黏膜具有一定的保护性作用,且疗效与剂量有关。
Objective To observe the change in gastric mucosa of the rats with acute cerebral infarction and study the protective effect of tongqiao huoxue decoction on gastric tissue in cerebral infarction at the acute stage. Methods The intraluminal middle cerebral artery occlusion with thread bolt method was used to prepare focal cerebral ischemia rat model. All of the rats were divided into a normal control group,a sham- operation group,a cerebral infarction group,a cimetidine group,a regular dose group and a low dose group of tongqiao huoxue decoction. The corresponding drugs were applied with gastric infusion in each group. Gastric infusion was given continuously,once a day and the rats were sacrificed in 4 days. The gastric tissue was separated and GAS was observed under electric microscope. Results In the cerebral infarction group,GAS was apparently higher than the other groups( P < 0. 01),GAS in the regular dose group of tongqiao huoxue decoction was lower than that in the low dose group( P < 0. 01) and that was lower much apparently than that in the cimetidine group( P < 0. 01). Compared with the normal control group and the sham- operation group,the structure of gastric mucosa was destroyed apparently. The destroyed tissue was alleviated in the regular dose group and the low dose group of tongqiao huoxue decoction,as well as the cimetidine group. The efficacy in the regular dose group was better than that in the cimetidine group. Conclusion Gastric mucosa in cerebral infarction at the acute stage is damaged apparently,which is closely related to the increasing of GAS. Tongqiao huoxue decoction protects gastric mucosa in acute cerebral infarction and the efficacy is relevant with the dose.
