杜仲对骨髓间充质干细胞增殖和向神经元分化的影响 被引量:11

摘要 目的探讨杜仲对骨髓间充质干细胞(BMSCs)增殖和向神经元分化的影响。方法自成年小鼠胫骨和股骨骨髓分离BMSCs并培养于含10%FBS的DMEM培养基中。将杜仲(10 mg/ml)加入含第4代BMSCs的培养基中,7 d后,通过相差显微镜观察和MTT法检测细胞增殖。28 d后,通过免疫荧光检测细胞分化,通过RT-PCR分析与细胞分化相关基因的表达。结果加入杜仲7 d后,实验组BMSCs的增殖能力提高了(32.2±1.86)%,与对照组比较,差异有统计学意义(P<0.01)。28 d后,3.77%BMSCs分化成为NF+神经元,5.09%的BMSCs分化成为MAP2+神经元。NF-H基因的表达,实验组明显高于对照组(P<0.05)。OCT-4基因的表达,实验组显著低于对照组。结论杜仲可提高BMSCs的增殖和向神经元分化的能力。 Objective To explore the impacts of eucommia on the proliferation of bone mesenchymal stem cells( BMSCs) and neuronal differentiation. Methods BMSCs were isolated from tibia and femur of adult mice and cultivated in DMEM medium containing 10% FBS. Adding eucommia( 10 mg / ml) into the medium containing the 4th generation of BMSCs,in 7 days,the cell proliferation was observed under phase contrast microscope and determined by MTT method. In 28 days,the cell differentiation was determined by immunofluorescent staining and the expressions of relevant genes were analyzed by RT- PCR. Results In 7 days after adding eucommina,in the experiment group,BMSCs proliferation was improved by( 32. 2 ± 1. 86) %,indicating the significant difference as compared with the control group( P < 0. 01). In 28 days,3. 77% of BMSCs were differentiated as NF+neurons and 5. 09% of BMSCs as MAP2+neurons. NF- H expression in the experiment group was higher apparently than that in the control group and OCT- 4 expression in the experiment group was lower significantly than that in the control group. Conclusion Eucommia increases BMSCs proliferation and the capacity of neuronal differentiation.
