摘要
为探究微生物发酵生产番茄红素的方法,以粗糙脉孢菌(Neurospora crassa)3.1607为发酵菌种,通过Box-Behnken原理进行响应面法优化发酵工艺条件。用研磨法结合超声波破碎法破碎菌体并以乙酸乙酯-丙酮(2∶1,V/V)混合有机溶剂浸提色素,对产物进行高效液相色谱分析,以乙腈-二氯甲烷(10∶90,V/V)为流动相并在472 nm波长处检测,能很好地分离出番茄红素。最终得到番茄红素最佳摇瓶发酵生产工艺为:发酵温度30℃、6.5%接种量、100 r/min振荡培养110 h,得到番茄红素产量达10.09 mg/L。
Fermentation conditions of Neurospora crassa for the production of lycopene were optimized by response surface methodology with Box-Behnken design. The cultured cells were disrupted by ultrasonication and milling for the extraction of pigment with ethyl acetate-acetone(2:1, V/V) before analysis by high performance liquid chromatography(HPLC). Good separation of lycopene was achieved by using acetonitrile-dichloromethane(10:90, V/V) as the mobile phase, and the detection was performed at 472 nm. The yield of lycopene was 10.09 mg/L under the optimal fermentation conditions: 30 ℃,an inoculum size of 6.5%, a shaking speed of 100 r/min and 110 h.
