沉默GRP94抑制宫颈癌细胞的侵袭和迁移 被引量:2

摘要 目的:探究GRP94在不同宫颈癌细胞株中的表达情况,及GRP94对细胞侵袭和迁移能力的影响。方法:采用Western blot和qRT-PCR检测不同类型的宫颈癌细胞株中GRP94的表达水平。选择高表达GRP94的细胞株,应用RNA干扰技术下调GRP94的表达,采用划痕实验和Transwell侵袭实验检测细胞侵袭和迁移能力的改变。结果:与正常的宫颈细胞相比,GRP94在不同的宫颈癌细胞株(C33A、CaSki、HeLa、HeLa229、MS751、ME-180、SiHa和HCC 94)中表达均升高,且GRP94在CaSki和MS751中表达量高,在ME-180、SiHa和HCC 94中表达量中等,在C33A、HeLa和HeLa 229中表达量低。选用GRP94高表达的CaSki细胞,下调GRP94的表达后,划痕实验和Transwell侵袭实验结果显示细胞的侵袭和迁移能力显著下降。结论:GRP94在宫颈癌细胞中高表达,沉默GRP94的表达能抑制宫颈癌细胞的侵袭和迁移能力。 Objective: To investigate the expression of GRP94 in different cervical cancer cell lines and its effect on the invasion and migration of cells. Methods: Western blot and qRT-PCR were performed to detect the expression level of GRP94 in different cervical cancer cell lines. The cell line with high expression of GRP94 was selected. The expression of GRP94 in selected cervical cancer cell line was knocked down by using RNA interference technique.The change of cells’ invasion and migration ability was detected by wound-healing assays and Transwell invasion assays. Results: The expression of GRP94 in all different cervical cancer cell lines( C33A,CaSki,HeLa,HeLa 229,MS751,ME-180,Si Ha and HCC 94) was higher than normal cervical cells,and it was high in CaSki and MS751,but moderate in ME-180,Si Ha and HCC 94,and low in C33 A,HeLa and HeLa 229. The CaSki was selected to knock down the expression of GRP94,wound-healing assays and Transwell invasion assays showed that the invasion and migration ability of cells decreased significantly after down regulated GRP94. Conclusion: GRP94 is highly expressed in cervical cancer cells. Silencing of GRP94 can inhibit the invasion and migration ability of cervical cancer cells.
