摘要
在制备多克隆抗体的基础上,研制用于检测莠去津残留的间接竞争ELISA试剂盒。结果表明:莠去津抑制率回归曲线为y=19.762x+5.6026,相关系数R2=0.981,线性检测范围为1~5000μg·L-1,最低检测限为5.35μg·L-1,IC50为176.44μg·L-1。莠去津多克隆抗体的交叉反应率小于20%,莠去津试剂盒的添加回收率均高于75%,供试样品检测结果的批内和批间变异系数均低于10%。试剂盒在4℃或-20℃下贮存270d仍有效。
Based on the preparation of polyclonal antibody against atrazine, an indirect ELISA kit was developed for detecting the residue of atrazine. Performance tests showed that the regression equation of inhibition rate for atrazine was y =19.762 x +5.6026(correlation coefficient, R2=0.981), with a linear detection range from 1 to 5000 μg·L-1. The indirect ELISA kit has an IC50 of 176.44 μg·L-1reaching a detection limit of 5.35 μg·L-1. The cross-reaction ratio of the kit for other triazines was less than 20%. The recovery rates of the kit for atrazine in different kinds of samples were higher than 75%. Both intra-assay and interassay coefficients of variation were less than 10%. The developed kit could be stored at 4℃ or-20℃ for a period of 270 days.
