清肺化浊行血宽中方对PM2.5引起慢性支气管炎大鼠肺组织病理学及血清IL-8、TNF-α的影响 被引量:27

摘要 目的应用鼻腔滴注法建立细颗粒物(PM2.5)致小鼠慢性支气管肺炎动物模型,以研究清肺化浊行血宽中方对改善PM2.5引起慢性支气管炎大鼠肺组织病理学及血清IL-8、TNF-α的影响。方法选取大鼠40只,雌雄各半,随机将其等量分为空白组、模型组、中药低剂量组、中药高剂量组4组。空白对照组小鼠不做处理,正常饲养,持续60 d;模型组、中药低剂量组、中药高剂量组小鼠则通过每日1次鼻腔滴注PM2.5悬液,剂量为40 mg/kg,持续滴注60 d,观察3组大鼠出现咳嗽、气喘、毛发枯槁、活动减少,造模成功。造模成功后,空白组、模型组给予0.9%Na Cl灌胃;中药低剂量组给予浓度为0.825 g/ml的清肺化浊行血宽中方灌胃;中药高剂量组给予浓度为1.650 g/ml的清肺化浊行血宽中方灌胃;各组灌胃频率均为1次/d,0.1 ml/10 g,持续15 d。15 d后制作病理切片,光镜下观察各组大鼠肺组织改变并监测血清IL-8、TNF-α含量的变化。结果与空白组比较,模型组血清IL-8、TNF-α含量差异具有统计学意义(P<0.05);治疗后,与模型组比较,中药高剂量、低剂量组血清IL-8、TNF-α含量均显著降低(P<0.05);病理学显示:中药高、低剂量组均对大鼠肺组织病理损伤有不同程度修复作用。结论清肺化浊行血宽中方对PM2.5引起的慢性支气管炎具有修复损伤肺组织、改善气体交换空间的作用,这可能与减少血清IL-8、TNF-α含量有关。 Objective To develop a rat model of particulate matter 2. 5 microns( PM2. 5) related chronic bronchitis,and study the effect of TCM Qing Fei Hua Zhuo Xing Xue Kuan Zhong prescription on lung histopathology and serum IL-8 and TNF-α levels in model rats. Methods Forty rats were randomly divided into control group,model group,low dose TCM group and high dose TCM group( 10 rats in each group). The rats in the control group did not receive any treatment for 60 days; the rats in other three groups received post nasal drip of PM2. 5 suspension( 40 mg/kg once a day) for 60 days to develop an animal model. Then,the control group and model group received intragastric administration of 0. 9% Na Cl,the low-dose TCM group received intragastric administration of TCM Qing Fei Hua Zhuo Xing Xue Kuan Zhong prescription liquid medicine at a dose of 0. 825 g/ml,and the high-dose TCM group received intragastric administration of TCM Qing Fei Hua Zhuo Xing Xue Kuan Zhong prescription liquid medicine at a dose of 1. 65 g/ml,once a day for 15 days. After the treatment,the morphological changes of lung tissue were observed under light microscope,and the levels of serum IL-8 and TNF-α were tested. Results The levels of serum IL-8 and TNF-α in the model group were statistically different from those in the control group( P < 0. 05). After the treatment,the levels of serum IL-8 and TNF both in the low-dose and high-dose TCM group were decreased as compared with the model group( P < 0. 05). The pathologic findings showed that the pathological damage of lung tissue was repaired at different levels both in the low-dose and high-dose TCM group( P < 0. 05). Conclusion TCM Qing Fei Hua Zhuo Xing Xue Kuan Zhong prescription can repair the damaged lung tissue and improve the space of gas exchange in rats with chronic bronchitis caused by PM2. 5,which may be related to the decrease of serum IL-8 and TNF-α.
