人参总皂甙对大鼠血浆皮质酮及肝糖原作用的昼夜节律

CIRCADIAN VARIATION OF GINSENG TOTAL SAPONINS ON PLASMA CORTICOSTERONE AND HEPATIC GLYCOGEN IN RATS

摘要 本文在生理性昼夜节律基础上,探索人参根总皂甙对雄性大鼠血浆皮质酮和肝糖原的影响。结果表明,用药后对于血浆皮质酮作用的强弱随昼夜的时辰而昇。08:00时最强,而在20:00和24:00时较弱。在同一时间(08:00时)取材,皂甙对皮质酮的作用存在剂量反应关系,但对肝糖原则无。文末对于皮质酮与肝糖原的关系和评价人参滋补作用的药理指标进行了讨论。 By applying a chronopharmacological approach, studies were carried out to determine the effect of total saponins extracted from radix ginseng (Panax ginseng C.A. Meyer; GTS) on plasma corticosterone and hepatic glycogen in male rats. Animals were acclimated to a 12hrs./12hrs. light-dark cycle, 22±2℃ at least for one week before the assay. Functional activities were assessed every 4 hrs. for 24 hrs.(1) In the physiological condition plasma corticosterone level and hepatic glycogen content displayed a stable circadian change which fit a single cosine curve. The peak value cccured at 20:00 hrs. for corticosterone and at 12:00 hrs.for glycogen, the trough value occured at 68:00 hrs. for corticosterone and 20:00 hrs. for glycogen.(2) After administration of GTS at the dose of 40mg/kg body Wt. (ip) the plasma corticosterone rose from 10.20±1.96 to 41.90±2.76μg/dl at 08:00 hrs., but this increase may change at other times according to circadian rhythm. (3) A dosage-dependent manner was found for plasma corticosterone but not for glycogen when rats were killed and determination was carried out in the morning. Linear relationship was obtained at doses from 0-40mg/kgo When GTS dosage exceeded the limit the pharmacological effects maintained at an identical level.It suggests that circadian rhythm may be an important factor in the pharmacological response to GTS.
