摘要 目的:观察年龄对成年小鼠嗅球细胞增殖与神经发生的影响。方法:用5-溴-2-脱氧尿核苷(5-bromo-2’-deoxyuridine,BrdU)标记新生细胞,免疫组化染色后统计新生细胞数量;应用BrdU与TuJ1/GFAP双标记,鉴别出BrdU阳性细胞的细胞类型。结果:老年组小鼠嗅球细胞增殖与神经发生明显低于青年组。结论:成年小鼠嗅球的细胞增殖与神经发生随着年龄的增长而减慢,这可能是嗅觉减退的结构基础之一。 Objective:To study the effects of age on cell proliferation and neurogenesis in the adult mouse olfactory bulb.Methods:Proliferating cells were labeled with5-bromo-2'-deoxyuridine(BrdU)for10days.BrdU immunohistochemistry was conducted to dertermine the number of newly generated cells and double immunolabeling undertaken to co-localize BrdU-positive cells with TuJ1(Tubulin-βAb-3,neu-ron-specific early differentiation marker)/GFAP(rabbit anti-glial fibrillary acid protein).Results:The number of new-born cells in the aged mice was significantly lower than that in the young mice.Conclu-sion:Cell proliferation and neurogenesis in the olfactory bulb decrease with the age increase of the adult mice,which may be related to the decrease of odor discrimination in the aged.
