褪黑素对化学诱变剂诱发微核形成的保护作用

摘要 目的 通过体内和体外实验观察褪黑素 (MT)对化学诱变剂诱发的人双核淋巴细胞微核和小鼠骨髓嗜多染红细胞微核形成的影响。方法  (1 )以丝裂霉素 (MMC)为阳性对照 ,观察用 3种浓度MT预处理人淋巴细胞的双核细胞微核率 ;(2 )以环磷酰胺 (CP)为阳性对照 ,分别用 3种剂量MT给小鼠灌胃 ,观察小鼠骨髓嗜多染红细胞的微核率。结果  (1 ) 0 0 1、0 1 0和 1 0 0mmol LMT +MMC处理组的双核淋巴细胞微核率与MT浓度呈负相关 ;(2 ) 0 1、1 0和 1 0 0mg kgMT +CP处理组的小鼠骨髓嗜多染红细胞微核率均呈剂量依赖性降低。 Objective To observe the effects of melatonin on micronucleus formation of binuclear cell in lymphocyte from human peripheral blo od and mouse bone marrow polycychromatic erythrocyte which induced by chemical mutagen.Methods (1)Lymphocytes were cultured in vitro in the presence o f mitomycin C(MMC)(positive control),and 0 01,0 10,1 00 mmol/L of melatomi n was added on in the treated groups.Binuclear lymphocytes were examined for the micronucleus rate.(2)The mice were administered with 0 1,1 0,10 0 mg/kg me latonin and cyclophosphamide(CP) while positive control only treated with CP.The rate of micronulclei of mouse bone marrow polycychromatic erythrocyte was exami ned.Results (1)The micronuleus rates of binuclear lymphocytes exhib ited negative correlation with the concentration of melatonin.(2)The micronuleus rates of mouse bone marrow polycychromatic crythrocytes which were exposed to 0 1,1 0,10 0 mg/kg melatonin with CP were obviously lower than that of posit ive control and its decrease was concentration-depended.Conclusion It seems that melatonin has obviously anti-mutagenic ef fect.
