游泳运动对高脂饮食老年大鼠胰岛素抵抗和血清瘦素的影响 被引量:11

Effect of swimming exercise on insulin resistance and serum leptin of elderly rats on high-fat diet

摘要 目的 研究单纯高脂饮食及不同强度运动对大鼠胰岛素敏感性和血清瘦素的影响。 方法  4 8只 15月龄雄性SD大鼠随机分为对照组、高脂组、高脂加 90min运动组及高脂加 4 5min运动组 ,两运动组大鼠在高脂饮食同时均进行 15周不同强度的游泳运动。实验结束测定大鼠血糖、胰岛素、胆固醇 (TC)、甘油三酯 (TG)、高密度脂蛋白胆固醇 (HDL C)、低密度脂蛋白胆固醇 (LDL C)和瘦素 ,计算胰岛素敏感指数 (ISI)和肥胖评定 (Lee)指数。 结果 高脂组ISI〔 - (6 6 3± 0 39)〕较对照组〔 - (5 5 7± 0 31)〕明显低 (P <0 0 1) ;而运动组ISI分别为〔 - (5 5 2± 0 2 8)和 - (5 36±0 2 9)〕 ,均较高脂组明显升高 (均P <0 0 1)。两运动组间ISI差异无显著性 (P >0 0 5 )。运动后两组血清瘦素分别为 (2 2 3± 0 84 )ng/ml和 (3 0 2± 1 39)ng/ml,较高脂组 (7 94± 3 6 7)ng/ml明显下降 (P <0 0 1)。血清瘦素与胰岛素呈正相关 (r =0 2 82 ,P <0 0 5 ) ;与ISI负相关 (r =- 0 4 2 3,P <0 0 1)。 结论 单纯高脂饮食可致大鼠胰岛素抵抗 ,适当运动可增加高脂饮食大鼠的胰岛素敏感性 ;血清瘦素水平与胰岛素、胰岛素抵抗相关。 Objective To investigate the effect of high-fat diet on insulin sensitivity and the effect of swimming exercise on insulin sensitivity and serum leptin in elderly rats. Methods Hyperlipidemia model in rats was established by feeding high-fat diet. Four experimental groups of rats were investigated: normal controls, high-fat diet group, high-fat diet and 90 minutes exercise group (90 min group); high-fat diet and 45 minutes exercise group (45 min group). Both exercise groups were swimming-trained for 15 weeks with high-fat diet. Serum glucose, insulin, total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C) and leptin were determined. Insulin resistance was evaluated with ISI, obese was evaluated with Lee index. Results ISI in high-fat diet group was decreased compared with that of the normal control group 〔-(6.63+ 0.39) vs -(5.57+0.31), P <0.01〕; there was no significant difference in both exercise groups(-5.52±0.28 vs -5.36±0.29, P >0.05); but in both exercise groups, the ISI was increased compared with high-fat diet group ( P <0.01); serum leptin levels (2.23±0.84 ng/ml and 3.02±1.39 ng/ml)in both exercise groups were significantly lower than that of the only high-fat diet group ( P <0.01)..There were positive correlation between serum leptin and insulin( r =0.282, P <0.05). Serum leptin level was negatively correlated with ISI( r =-0.423, P <0.01). Conclusions High-fat diet results in IR in rats. Appropriate exercise may improve the insulin sensitivity in high-fat diet rats. There is a close relationship between insulin, IR and leptin.
