摘要
目的 :观察家兔颈椎间盘退变过程中血液流变学、髓核蛋白多糖、胶原及椎间盘病理的变化和丹参注射液对这些变化的影响。方法 :通过破坏家兔颈推动、静力平衡 ,建立家兔颈椎间盘退变的模型。 6 0只兔子随机分成 3组 ,颈椎间盘退变组 (A) 4 0只 ,分 4个亚组 (A1、A2、A3、A4 ) ,即为造模后 1、3、5、7月组 ,每组各 10只 ;退变加丹参治疗组 (B) 10只 ,造模后饲养 6个月后 ,每日腹腔内注射丹参注射液 2ml ,1个月后处死 ;正常对照组 (C) 10只。通过测定血液流变学、髓核蛋白多糖、胶原的含量 ,比较各组之间的差异 ,在光镜、电镜下观察椎间盘组织的细胞、基质形态及超微结构的变化。结果 :丹参治疗组 (B)家兔全血高切黏度、低切黏度、血沉与A4组比较P <0 0 1,红细胞压积同A4组比较P <0 0 5 ,以上均有显著性差异 ;血浆黏度、血浆纤维蛋白原同A4组比较 (P >0 0 5 ) ,无显著性差异。髓核蛋白多糖、胶原与A4组比较 (P <0 0 1) ,均有显著性差异。结论 :丹参能改善家兔血液流变学和微循环的灌注 ,增加椎间盘髓核的蛋白多糖 ,降低胶原的含量 ,促进组织修复、再生 ,能够延缓和抑制颈椎间盘的退变。
Objective:Observe the change of hemorheology、nucleus pulposus of cervical intervertebral disk proteoglycan、 collagen and pathology in the process of rabbit's cervical intervertebral discs deterioration, and impacts salvia miliorrihizae injection has on those changes.Methods: set up the model of rabbit's cervical intervertebral discs deterioration by destroying rabbit's dynamostatic balance. Sixty rabbits were at random divided into three groups.In the cervical intervertebral discs deterioration group(A) there were forty rabbits, which were divided into four subgroups(A1、 A2、 A3、 A4),i.e. after setting up the model month 1、 month 3、month 5、month 7 group in which there were ten rabbits respectively;Group(B):there are ten rabbits with cervical intervertebral discs deterioration but treated with salvia miliorrihizae Injection after setting up the model these rabbits were raised six months, every day they were injected salvia miliorrihizae Injection 2ml into their abdominal cavity,they were executed one month later;there were ten rabbits in the normal contrast group(C).We measured hemorheology, proteoglycan and collagen in nucleus pulposus, compared the differences between the groups.Through optical microscope and electron microscope to observe the change of cell of intervertebral disk tissue and matrix form and ultrastructure. Results: Compared B group to A4 group, it was P <0 01 in the terms of the whole blood viscosity(bvh,bvl) and ESR,it was P <0 05 in terms of HCT,all the above comparisons indicated significant differences,in terms of plasma viscosity(pv) and fibrinogen(FB),it was P >0 05,which indicated no significant difference.In terms of intralaminar nuclei proteoglycan and collagen,it was P <0 01, which indicated significant differences.Conclusion: Salvia miltiorrhizae can improve rabbit's hemorheology and irrigation of micro circulation, enhance the percentage of intervertebral discs intralaminar nuclei proteoglycan, lower the percentage of collagen, promote tissue's repair and reproduction, put off and hinder retrogression of cervical intervertebral discs.
