摘要
北美红杉种子经消毒处理后接种到MS培养基中所获得的无菌外植体茎段,在MS+6 BA1.0mg/L培养基中,诱导出愈伤组织和芽;在MS+6 BA0.1+KT0.3+NAA0.1培养基中继代培养,扩增倍数(MR值)约5~12倍;将继代培养芽丛中每具两个节的芽茎段接种到生根培养基MS(3/4NH4NO3,3/4KNO3)+KT0.2+NAA0.05+IBA0.2+VB20.2+B90.3中,50d左右生根率达65%;练苗30~40d后移栽,成活率65%以上。
The stems of the aseptic explants which the disinfected seeds of Sequoia sempervirens were inoculated on MS culture medium to produce were induced to produce callus and buds on the culture medicum of MS+6BA1.0 mg/L. These callus and buds were secondary cultured on the medium of MS+6BA0.1+KT 0.3+NAA0.1 with the multiply rate(MR)of 5-12. The stems of buds with two joints were inoculated on MS(3/4NH4NO4,3/4KNO3)+KT0.2+NAA0.05+IBA0.2+VB20.2+B90.3 and the rate of rootage reached 65% about fifty days later. The survival ratio of transplanting was beyond 65% after 30~40 days' seedling training.
