摘要
目的 研究顺铂 (cisplatin ,CDDP)和 5 氟脲嘧啶 (5 fluorouracil,5 FU)联合化疗对口腔鳞癌 (Tca8113细胞株 )的体内外抗瘤作用。方法 应用CDDP和 5 FU单独或联合治疗口腔鳞癌细胞 (Tca8113细胞系 )及其裸鼠移植瘤 ,采用MTT法检测其体外杀伤作用 ;观察移植瘤的生长情况及组织病理学改变 ;高效液相色谱法 (HPLC)检测血液及移植瘤内 5 FU的浓度。结果 CDDP和 5 FU均对口腔鳞癌细胞具有强的体外杀伤作用 ,其IC50 分别为 :0 5 9μmol/L和 2 33μmol/L。与对照组比较 ,移植瘤经CDDP和 5 FU单独或联合治疗后生长均明显受到抑制 (P <0 0 0 1) ,其抑瘤率分别为 :5 7 0 % (5 FU)、84 4 % (CDDP)和 97 2 % (5 FU +CDDP) ,CDDP和 5 FU存在协同的抑瘤作用(P <0 0 0 1) ;组织病理学显示各治疗组移植瘤内出现囊性变和灶性坏死 ;肿瘤组织和血液中可检测到高浓度的 5 FU。结论 CDDP和 5
Objective To investigated the anticancer effect of cisplatin (CDDP) and 5-fluorouracil (5 FU) combinative chemotherapy in oral squamous carcinoma cell line (Tca8113 cell line). Methods Tca8113 cell line and its xenografts in BALB/c nude mice were treated with CDDP and/or 5 FU. The cytotoxicity on Tca8113 cells was measured by MTT assay. The growth state of Tca8113 cell xenografts and its histopathological changes after treatment were observed. High performance liquid chromatography (HPLC) was performed to determine the concentration of 5 FU in serum and tumor tissue in vivo. Results CDDP and 5 FU had strong cytotoxicity on Tca8113 cells, the IC 50 was 0 59μmol/L and 2 33 μmol/L respectively. Compared to the control group, mice treated with CDDP or/and 5 FU demonstrated significant tumor regression ( P <0 001); the tumor inhibition rate was 57 0%(5 FU), 84 4%(CDDP), 97 2%(5 FU+ CDDP), respectively. There had cooperative anticancer effect between CDDP and 5 FU by factorial experiment. Pseudocysts and necrotic morphology were found in all treatment groups. There had high concentration of 5 FU in tumor tissue and blood. Conclusion The combinative chemotherapy of CDDP and 5 FU is an effectively chemotherapy regiment for oral squamous cell carcinoma.
