缺碘大鼠补充不同剂量碘酸钾和碘化钾对甲状腺和血液抗氧化能力的影响 被引量:15

摘要 目的 观察缺碘Wistar大鼠补充不同剂量碘酸钾(KIO_3)和碘化钾(KI)两种碘剂治疗缺碘性甲肿时,甲状腺抗氧化能力的变化。方法 在成功复制缺碘性甲状腺肿动物模型的基础上,分别给予适量碘(1倍)、5倍和50倍于适量碘的KIO_3和KI这6种措施进行干预治疗,治疗1月和3月后,测定血和甲状腺组织匀浆中的超氧化物歧化酶(SOD)活力以及丙二醛(MDA)含量。结果 (1)甲状腺组织抗氧化能力:①6个治疗组的SOD和MDA均明显低于低碘组(LI组),但总体趋势仍高于正常对照组(NI组);②不同补碘剂量之间的比较:SOD和MDA在治疗1月时均随剂量加大呈下降趋势,但在治疗3月后KIO_3组两个指标均随剂量加大呈升高趋势,KI组未发现增高或降低趋势;③不同补碘剂型之间的比较:KIO_3和KI组在三种碘水平上比较发现,治疗1月时,KIO_3组的SOD均高于KI组,MDA与KI组比较也似有增高趋势;治疗3月后,KIO_3组的SOD与KI组比较似有降低趋势,但MDA似有升高趋势。(2)血抗氧化酶:①LI组与NI组的SOD和MDA均无差异;②与LI和NI组比较,除1×KI外,其余5个治疗组的SOD均与LI和NI组无差异。KIO_3组MDA均低于NI组,1×KIO_3同时还低于LI组,而KI组与LI和NI组均无差别;③KIO_3组和KI组的SOD均随补碘剂量增大似有降低趋势,但MDA变化不明显;④在三种补碘水平上。 Objective To study the effect of different dosage KIO3 and KI on thyroid antioxidative capability in goiter induced by iodine deficiency. Methods The rats with iodine deficient goiter were divided into 6 groups and given by different dosage of KIO2 and KI for 3 months respectively, that was normal iodine (1 fold ) ,5 fold and 50 fold KIO3 and KI.The other 2 groups of low iodine(LI)and normal iodine(NI)remained as the control. 1 month and 3 months later, the rats were sacrificed.Serum and thyroid SOD activity and MDA content were measured. Results (1)The capability of thyroid antioxdition ①The SOD and MDA in 6 treated groups were markedly lower than LI group and still higher than the NI group. ② After treatment for 1 month, the more iodine was given, the lower SOD and MDA were detected, but no much change was found in 3 dosage KI groups. ③ After treatment for 1 month, SOD of KIO3 groups were higher than that of KI groups, but after treatment for 3 months, SOD of KIO3 groups might lower than that of KI groups. MDA of KIO3 groups might higher than that of KI groups at 1 and 3 month's treatment. (2)The capability of serum antioxdition ①There was no difference in SOD and MDA between LJ and NI groups. ②SOD in the other 5 groups, except 1 fold KI group , was similar to LI and NI group. MDA of 3 different dosage KIO3 groups were all lower than that of NI group. Furthermore, MDA of 1 fold KIO3 group was also lower than that of LI group. But MDA of KI group was similar to LJ and NI groups. ③The more iodine was given, the lower SOD in KIO3 and KI groups was found , however, MDA in 6 treatment groups didn' t change with the dosage increase of iodine intake. ④SOD and MDA of KIO3 groups were lower than those of KI groups at 3 iodine - supplied levels. Conclusions (1) Different dosages of KIO3 and KI were all effective for recovering antioxidative capability in iodine deficient rats ,but the recover procedure might need a long time . (2) High dosage of KIO3 and KI given for a short time (1 month) might have a good effect on thyroid antioxidation, but it was not good to give such high dosage of iodine for a long time. (3)KIO3 in appropriate dosage had a quite good effect on thyroid antioxidation compared to KI even given for a long period. (4)Blood had a stronger antioxidative and compensative capabilities than the thyroid gland. (5)These findings above demonstrated that KIO3 in approriate dosage was safe and effective for correction of iodine deficiency.
