摘要
目的 探讨应用大剂量异丙基肾上腺素 (ISO)诱导大鼠心力衰竭模型的剂量和时限 ,以及如何提高动物存活率和模型成功率。方法 用不同剂量 (85mg kg ,170mg kg,340mg kg)ISO对大鼠进行皮下注射 2次 ,分别在不同时间段进行超声心动检查 ,测量左室舒张末径 (LVEDD) ,左室收缩末径 (LVESD) ,射血分数 (EF)。以EF <4 5 %作为心衰标准 ,来评估和鉴定模型的成功率 ,确定ISO应用的剂量和时限。结果 随着剂量的增大 ,模型成功的时间逐渐缩短 ,动物死亡率增加 (P <0 .0 1) ;随着时间的延长 ,动物心衰程度逐渐加重 ,死亡率增加 (P <0 .0 1) ;随着经验的积累 ,动物的急性死亡率可以降低。结论 ISO所致的心力衰竭有剂量和时间依赖性 ,采取必要的措施可以降低动物死亡率 ,提高模型成功率。
Objective To explore the dosage and time limit in application of isoproterenol(ISO) in rats with ISO-induced congestive heart failure(CHF) and h ow to improve the survival rate and successful rate of animals. Methods Rats received two subcutaneous injection of ISO which were separated by a 24-interval in different dosage.Cardiac function was assessed by echocardiogram at different time.The indexes include left ventricular (LV) end diastolic diameter (LVEDD),LV end systolic diameter (LVESD) and ejection fractio n (EF).The standard of CHF is EF<45%. Results Along with increased doses of ISO there was a decrease of t ime limit of a successful model,and an increase of mortality (P<0.01).As tim e go es on,the degree of CHF became more serious (P<0.01),the mortality was incre asing. Conclusion High doses of ISO produce a time-and dose-dependent impairment of cardiac function in CHF.And the acute mortality for the rat model of CHF is significantly improved with technical training and increasing of expe riences.
