摘要
目的:进一步探讨姜黄素对LPS诱导的系膜细胞单核细胞趋化蛋白-1(MCP-1)表达的影响。方法:采用不同浓度姜黄素处理体外培养的人肾小球系膜细胞,分别收集上清液及细胞,应用ELISA的方法测定上清液中MCP-1蛋白表达量,逆转录多聚酶链反应(RT-PCR)方法测定系膜细胞MCP-1基因的相对表达量。结果:LPS可显著上调系膜细胞MCP-1基因表达,同时刺激系膜细胞分泌MCP-1蛋白;姜黄素则呈浓度依赖性地抑制LPS诱导的系膜细胞MCP-1基因及蛋白表达。结论:姜黄素能抑制LPS诱导的肾小球系膜细胞MCP-1基因和蛋白的表达。
Objection:It has been shown curcumin inhibit the human mesangial cell growth and decrease its extracellular matrix release. This study was to determine whether curcumin inhibits the monocyte chemoattractant protein-1 (MCP-1) gene expression and MCP-1 protein release in human mesangial cells. Methods: Human mesangial cells were cultured with or without lipopolysaccharide (LPS,10 (Jig/ml) in presence or absence of various concention of curcumin (4, 16, 200 u,mol/L), collected the supernatant and the cells respectively. The levels of the MCP-1 protein in the supernatant were determined by means of ELISA after subconfluent quiescent mesangial cell incubated with various concentration of curcumin for 24 hours in vitro. And the MCP-1 mRNA expression were measured by semi-quantitative RT-PCR. Results: LPS could upregulate the MCP-1 mRNA expression and induce the MCP-1 protein release in human mesangial cells. Whereas curcumin could downregulate the mRNA expression and protein production of MCP-1 in mesangial cell induced by LPS in a dose-dependent manner. Conclusion: Curcumin could inhibit secretion of MCP-1 in mesangial cells induced by LPS and downregulate the mRNA expression of MCP-1 at the same time. The results suggest that curcumin may be profitable in curbing the inflammatory cells infiltrating in renal tissue and might be beneficial in retarding progressive development of glomerulonephritis.
