摘要
目的 探讨日本血吸虫TPI DNA疫苗对血吸虫肝脏虫卵肉芽肿的免疫调节作用,以评价其作为抗病疫苗的潜能。方法 30头松江猪分为3组。A组(TPI组)每头猪于两侧臀部肌注500μgpcDNA3.1-SjCTPI质粒DNA;B组(TPI+IL-12组)每头猪肌注500μg pcDNA3.1-SjCTPI DNA疫苗及500μg pcDNA3.1-P35和500μg pcDNA3.1-P40质粒DNA的混和物;C组(对照组),每头猪肌注500μg pcDNA3.1质粒DNA。于第0、3、6周共免疫3次。末次免疫后30d,每头猪采用背部贴片法攻击感染600条日本血吸虫尾蚴,攻击后45 d剖杀实验猪,取肝脏,于同一部位切取1.5 cm^3的肝组织。按常规法制备石蜡切片。切片在NYD-1000型图像分析系统下观察,测定所有肉芽肿的面积和直径,并比较。结果 与对照组相比TPI组及TPI+IL-12组肉芽肿内炎性细胞明显减少。肉芽肿平均面积较对照组分别减少41.95%和42.50%,肉芽肿直径分别减少25.14%和25.66%。结论 SjCTPI DNA疫苗具有下调血吸虫肝脏肉芽肿的作用,可望成为一种血吸虫病抗病疫苗。
Objective To explore the effect of SjCTPI DNA vaccine on regulation of hepatic granuloma of schistosomiasis japonica and assess its potential to be as an anti-pathology vaccine. Methods Thirty Songjiang native pigs were divided into A,B,C groups. In group A (TPI group), each pig was immunized (i.m.) with 500 μg SjCTPI UNA in both buttocks. In group B (TPI+IL-12 group), each pig was immunized with 500μg SjCTPI plasmid DNA and the mixture of 500μg pcI)NA-P35 and 500μg of pcDNA P40 plasmid DNA by i.m. Each pig was immunized (i.m.) with 500μg of pcDNA3.1 as control in group C. Immunized for 3 times at week 0, 3, 6. At day 30 after last immunization each pig was challenged with 600 cercarae of Schistosoma japonicum and 45 days later the pigs were sacrificed.1.5 cm3 of liver piece was cut from each leaf of the liver. The paraffin sections were prepared by the routine procedure.The egg granulomas of liver in each section were investigated. The square measure and diameter of each granuloma were measured with NYD-1 000 Image Analysis System. The average square measure size and diameter of granuloma were calculated for each group and compared between groups. Results Compared with control group, the aver-age square measures of TPI group and TPI + IL-12 group were reduced by 41.95% and 42-50% respectively, the diameters were diminished by 25.14% and 25.66% respectively.Conclusion SjC TPI DNA vaccine could be down regulation of liver granuloma of schistosomiasis japonica and it could be potential as an anti-pathology vaccine of schistosomiasis.
