摘要 目的 观察丹酚酸B(SA B)对肝星状细胞 (HSC)分泌转化生长因子 β1(TGFβ1)与TGFβ1促HSC分泌胶原的影响。方法 用ELISA法测定培养 4d的原代与传代HSC的TGFβ1分泌量 ;添加TGFβ1于原代HSC中温育 2 4h后 ,[3 H]脯氨酸掺入与胶原酶消化法观察TGFβ1促进HSC分泌胶原的作用 ;Western印迹法检测HSCⅠ型胶原的表达 ;添加SA B ,观察对上述实验的干预。结果 传代HSC分泌TGFβ1的量明显高于原代HSC ;添加SA B后 ,对原代HSC的TGFβ1分泌量无明显影响 ,而传代HSC的TGFβ1分泌量显著减少。TGFβ1可显著促进HSC胶原的分泌和Ⅰ型胶原的表达 ,而SA B可拮抗TGFβ1的效应。结论 SA B可抑制传代HSC的TGFβ1分泌与减弱TGFβ1促HSC胶原的合成 ,这两个环节可能是SA B抗肝纤维化的主要作用机制。 Objective To observe the influence of Salvianoic acid B(SA-B) on secretion of transforming growth factor-β 1(TGF-β 1) by hepatic stellate cells(HSC) and to investigate the collagen secretion stimulated by TGF-β 1. Methods The secretion of TGF-β 1 in primary and activated HSC were assayed by ELISA. The collagen secretion stimulated by TGF-β 1 was detected by [ 3H] proline incorporation and collagenase digestion, and collagen I expression was by Western blot. The SA-B was incubated with HSC and its effects on the collagen production and TGF-β 1 secretion were observed.Results The secretion of TGF-β 1 in activated HSC was much higher than that in the primary HSC. SA-B could markedly inhibit the secretion of TGF-β 1 in activated HSC and TGF-β 1 increased the collagen secretion and collagen I expression significantly.Conclusion SA-B can inhibit the secretion of TGF-β 1 in activated HSC, and decrease the collagen synthesis of HSC stimulated by TGF-β 1.
