环氧化酶-2在乳腺癌中的表达及其与血管生成的关系 被引量:3

The relationship between cyclooxygenase-2 expression and tumor angiogenesis in human breast cancer

摘要 目的 观察环氧化酶 2 (cyclooxygenase 2 ,COX 2 )在乳腺癌中的表达 ,探讨其对血管生成的影响。 方法 取1996年 2月至 1998年 3月在我院行乳腺癌改良根治术或根治术患者 80例术后石蜡包埋组织。应用免疫组织化学方法检测了COX 2和血管内皮生长因子 (vascularendothelialgrowthfactor,VEGF)在乳腺癌中的表达 ,通过CD34单抗标记检测肿瘤微血管密度 (microvasculardensity ,MVD)。结果 COX 2和VEGF表达明显相关 ;COX 2和VEGF表达均与MVD相关 ;COX 2在 5 7.5 % (46 / 80 )乳腺癌中过表达 ,VEGF在 5 5 .0 % (44 / 80 )乳腺癌中过表达 ,与正常组织相比均具有显著差异。结论 COX 2与乳腺癌血管生成有关 ,VEGF可能是COX Objective To study the relationship between cyclooxygenase-2 (COX-2) expression and tumor angiogenesis in human breast cancer. Methods Archival primary breast carcinomas (n=62), adjacent DCIS (n=13) and DCIS alone (n=5) were analyzed for COX-2 and vascular endothelial growth factor (VEGF) expressions by immunohistochemistry using specific monoclonal antibodies. We also examined the microvascular density (MVD), using CD34 staining. Results A significant correlation was found between COX-2 and VEGF expressions (P< 0.05). Both COX-2 and VEGF were significantly correlated with MVD. COX-2 and VEGF genes were overexpressed in tumor specimens as compared with normal epithelia. Conclusion COX-2 is related to tumor angiogenesis in breast cancer. It is likely that VEGF is one of the most important mediators of the COX-2 angiogenic pathway.
