摘要
目的 探讨培哚普利 (perindopril)对链脲佐菌素 (STZ)诱导的糖尿病大鼠心脏的保护作用。 方法 雄性SD大鼠随机分为正常对照组 (NC)、DM对照组 (DC组 )、DM治疗组 (DP组 ) ,DP组每天给予培哚普利 2mg/kg。于病程 4、8、16周测血糖、心肌AngⅡ ,左室重量指数 (LVMI∶LVM/BW )并取左室心肌组织作电镜观察。结果 4、8、16周DC组LVMI均明显高于NC组 (P <0 0 1) ;而DP组LVMI明显低于DC组 ,第8、16周 ,DC组心肌AngⅡ明显高于NC组 (P <0 0 1) ,第 16周DP组心肌AngⅡ则明显低于DC组 (P <0 0 5 )。电镜观察表明 ,4周起DC组大鼠心肌开始有线粒体数目增加和肌原纤维灶性变性、溶解 ,随病程延长 ,病变加重 ,但DP组病变轻微。结论 DM大鼠有心肌局部RAS激活和超微结构改变 ,培哚普利能逆转或阻止这种改善 。
Objective To investigate the cardial protective effect of perindopril in diabetic rats induced by streptozotocin.Methods Male SD rats were randomly divided into normal control group(NC),diabetic control group(DC),diabetic group treated with perindopril(DP)(2 mg·kg -1 ·d -1 ).At 4,8,16 weeks,rats were respectively measured blood glucose,cardiac AngⅡ level,left ventricalar mass index(LVMI) and observed left ventricalar myocardium under the electon microscope.Results At 4,8,16 weeks,LVMI of DC groups were elevated statisically compared with NC group(P<0 01);While LVMI of DP groups were singnificantly declined at 8,16 weeks,cardiac AngⅡ were increased significantly in DC groups as compared with NC group(P<0 01) and group(P<0 01) and was obviously declined in DP group at 16 weeks(P<0 05).The electron microscopic findings showed the increase in amounts of mitochondria and myofibrosis and dissolution in the diabetic rats,with progression of the course,myocardial ultrastructural alteration in diabetic rats became more severe,while alteration in DP group were markedly improved as compared with DC groups.Conclusion There were activated cardiac RAS and myocardial ultrastructural alteration in diabetic rats,perindopril could reverse or prevent this alteration and may have protective effect on myocardium in diabetic rats.
