钙化是生物瓣原发性失功的主要原因。文章阐述了硅胶羟基乙叉二磷酸盐(EHDP)缓释块的制作,观察其释放和抗钙化的效果。EHDP缓释快体外第一个月释放较快,以后以20μg/日速度持续释放。对埋植于大鼠皮下的牛心包有抑制钙化作用。60天后实验组牛心包钙含量为14.3m/mg对照组为180.9μg/ml(P<0.01)。取出埋植的缓释块在体外也有EHOP释出。组织学检查大鼠股骨无骨骺增宽、钙沉积不良等副作用。
Calcification is the main cause of bioprosthetic heart valve failure. This investigation described the method of preparation, in vitro release and in vivo anticalcification efficacy of the slowly releasing silicone matrix bloxks containing disodium 1,1-hydroxyethelidene diphosphonate (Na_2EHDP) and dicalcium 1,1-hydroxyethelidene (Ca_1EHDP). The blocks can constantly release Na_2EHDP 20 μg/d afater rapid release in the first month in vitro and inhibit calcification of the subdermal bovine pericardial implants in rats for 60 days. The calcium level in the experimental group avereged 14.3 μg/mg, and in the control group 180.9μg/mg(P<0.01). The slowly releasing EHDP blocks removed from 60-d rat subdermal implants also demostrated EHDP release in vitro. Examination of the rat femur did not show any adverse effects such as epiphyseal thickening and calcium deposition abnormalities.
