摘要
在香菇种间原生质体融合中,对虎皮香菇进行失活原生质体标记,对香菇进行温度敏感型标记,分别以虎皮香菇和香菇的单核和双核体为亲本,用化学融合方法,以高温(35℃)为选择压,筛选并构建了32个不同组合的香菇种间融合子。这些融合子与亲本菌株形成明显的拮抗反应,在木屑代料培养基上产生形态各异的子实体,并形成了新的遗传性状——菌丝体无锁状联合,但能形成子实体。
The monokaryon and dikaryoh of Lentinus edodes and L. tigrinus are used as parents in interspecies protoplast fusion. The Protoplast of L. tigrinus is inactived with chemical method, and the mark of L. edodes is temperature sensitive. After fusion by PEG, 32 fusion products from different combinations were constructed under selection pressure of high temperature (35℃). These fusion products showed clearly antagonistic reactions with their parents, and produced morphologically different fruit bodies in sawdust substrate. A new genetic character of fusion products is that the mycelia can form fruit bodies without clamp connection.
