摘要
目的 采用体外细胞培养技术 ,模拟体内肿瘤转移过程 ,研究c9,t11-共轭亚油酸 (CLA)对肿瘤细胞侵袭及侵袭相关特性的影响。方法 用 0 ,2 5,50 ,10 0和 2 0 0 μmol/LCLA处理SGC -790 1人胃腺癌细胞 2 4h后 ,分别进行重组基底膜侵袭试验、趋向性运动实验和粘附试验研究CLA对侵袭、运动和粘附能力的影响。结果 在 2 0 0 ,10 0和 50 μmol/L浓度时 ,c9,t11-CLA对肿瘤细胞侵袭重组基底膜的抑制率分别为 53 7% ,40 9%和 2 9 3 %。同时 ,c9,t11-CLA处理后的肿瘤细胞与细胞外基质的粘附能力降低 ,但对肿瘤细胞趋化运动能力的影响不明显。结论 c9,t11-CLA可降低肿瘤细胞的体外侵袭能力 。
Objective To investigate the effects of c9,t11-conjugate d linoleic acid on cancer invasion and invision-related characteristics the met astasis cascades of tumor cells were imitated in cell culture.Methods The five levels of CLA in medium were designed as 0,25,50,100 and 200?μmol/L.After human gastric carcinoma cell line SGC-7901 were treated with c9,t11-CLA for 24?h,reconstituted basement membrane invasion assay and migration assay was used to evaluate invasive ability and migration a bility of cancer cells,adhesion assay was investigated the ability of treated ce lls to adhere to intracellular matrix.Results At the concentrations of 200,100 and 50?μmol/L, c9,t11-CLA suppressed the reconstituted basement memebrane invasion of SGC-790 1 by 53 7%,40 9% and 29 3%.Meanwhile,c9,t11-CLA could inhibit the adhesion t o laminin,fibronectin and matrigel.c9,t11-CLA did not,however,significantly inf luence the migration ability of treated cells.Conclusion c9,t11-CLA could suppress invasion of SGC-79 01 cells and change some invasion-related characteristics.
