摘要
目的 用Cocktail探针药物法 ,研究连翘对大鼠肝CYP4 5 0的影响。方法 将SD♂大鼠随机分组 ,给予连翘的水提物 ,以生理盐水组为空白对照 ,通过HPLC检测Cocktail探针药物的代谢率来评价各组CYP4 5 0。结果 给予连翘组的大鼠CYP1A2的水平显著低于对照组 ,CYP3A4的水平显著高于对照组 ,CYP2E1的水平没有显著变化。结论 连翘对大鼠CYP1A2有抑制作用 ,对大鼠CYP3A4有诱导作用 ,但对大鼠CYP2E1的影响不显著。
OBJECTIVE: To study the change of the hepatic CYP450 activity following administration of Fisch Fructus Forsythiae by Cocktail probe drugs in rats. METHODS: The rats were randomly divided into two groups. One group of rats were given Fisch Fructus Forsythiae plant extracts once daily orally for six days. Another group received orally normal saline once daily as the blank control group. The plasma concentrations of Cocktail probe drugs were determined by HPLC. RESULTS: The CYP1A2 content after received Fisch Fructus Forsythiae plant extracts was reduced, but the CYP3A4 content was enhanced. At the same time the CYP2E1 content was not markedly changed. CONCLUSION: It suggested that Fisch Fructus Forsythiae tended to be the inhibitor of the CYP1A2 and inducer of CYP3A4. But the CYP2E1 was hardly affected.
