摘要
目的:探讨免疫球蛋白对癫癎大鼠模型的神经细胞保护作用。方法:用印防已毒素(PTX)制备癫大鼠模型,在发作前、后分别给予免疫球蛋白进行预处理和治疗,用原位末端标记法及免疫组织化学法分别检测大鼠海马神经细胞凋亡、P53蛋白表达和大鼠脑组织匀浆的白细胞介素-2、4(IL-2,IL-4)的浓度变化,并设立对照组进行比较。结果:经免疫球蛋白预处理和治疗后的大鼠的神经细胞凋亡数量和P53蛋白表达均少于对照组(P<0.05),脑组织匀浆中IL-2,IL-4的浓度也低于对照组(P<0.01)。结论:免疫球蛋白能有效地治疗癫,可能是通过阻止神经细胞凋亡、减少P53蛋白表达和降低脑内免疫反应强度,达到神经细胞保护作用来实现的。
Objective:To determine whether immunoglobulins have the protective effects on the neurons in the epilepsy Wistar rat models.Methods:Rat models are induced by Picrotoxin(PTX Sig-ma),and were pre-treated and treated with immunoglobulins before and after seizures induced by PTX.Terminal deoxynucleotidyl thransferase-mediated dUTP-biotin in situ nick end labeling(TUNEL)technique and immunohistochemical method were used to detct number apoptotic neurons,expression of P53-protein in the hippocampus area,and the changes in the concentration of interleukins-2and-4in rat brain-homgente.The results were compared with those obtained in a control group.Results:In the group,the number of apoptotic neurons was smaller (P<0.05),and the expression of P53-protein and the con-centration of IL-2and IL-4were also lower than those in the control group(P<0.01).Conclusion:It is suggested that it is immunoglobulins exert a neuro-protective effect to inhibit the seizures induced by PTX through the protection against neuronal apoptosis,the reduction of expression of P53-protein and downregulation of the intensity of immunoreaction on that.
