网纹瓜(Cucumis melo Var.reticulatus)是名贵的优质甜瓜,为葫芦科、甜瓜属植物。我国引种后生长良好,产量高,经济效益好,每年从日本购进大量杂交种子。但由于种子不纯,基因型差异较大,植株不整齐,产量不稳定,而且价格昂贵。因此。
The paper reports the establishement of an autosomaclonc of Cucunis melo var. reticulatus in single cell culture, including callus induction from plumular axis and cotyledon, cell suspension culture and isolation of single cell and so on. The callus are reformed from the single cells in solidliquid dual layer MS medium with BA_(1.0), and then arc transferred to the MS solid medium contanning BA_(3.0). Some of these calli differentiated into buds, which formed roots in ^(1/2)MS medium supplemented KT_(1.0). 16 embryo cell clones with high ability of regeneration are selected from many clones, A large number of plantlets are obtained from the embryo clones.
The experiment showed that the solidliquid dual layer culture can obtain the highest plating efficiency of single cells and differentiation frequency of callus in three culture methods.
