摘要
目的 探讨氧化低密度脂蛋白 (OX -LDL)、理气活血方和方中理气、活血药对不同组分对人脐静脉内皮细胞 (HUVEC)生长活性的影响。方法 体外培养HUVEC ,将OX LDL与HUVEC共同孵育 ,模拟OX LDL损伤内皮细胞 (EC)的体内过程。理气活血方和川芎、赤芍药对 (芎芍药对 )及柴胡、枳壳药对 (柴枳药对 )组分分别与OX LDL损伤的HUVEC共同孵育 ,用四唑蓝比色试验检测法(MTT)检测细胞生长活性。结果 HUVEC和OX LDL共同孵育 6、12h和 2 4h后 ,HUVEC生长活性都受到不同程度抑制 ,和正常培养的HUVEC有显著差异 (P <0 0 5 )。理气活血方全方及方中柴枳药对、芎芍药对组分能提高OX LDL损伤HUVEC生长活性 (P <0 0 5 )。结论 理气活血方不同组分可以提高OX LDL损伤HUVEC的生长活性 。
Objective To investigate the effects of oxidized low density lipoprotein (OX-LDL), Liqihuoxue Formula (LF) and its different ingredient pairs on the activity of the growth of human umbilical venous endothelial cells (HUVECs). Methods First, HUVECs were incubated with OX-LDL to simulate the process in vivo of the OX-LDL injury of endothelial cells; and then, the OX-LDL-damaged HUVECs were incubated respectively with LF, the ingredient pair of Ligusticum chuanxiong and Paeoniae lactiflora (IP-LP), and the ingredient pair of Bupleurum chinense and Citrus aurantium (IP-BC). MTT assay was used to detect the activity of HUVECs growth. Results The activity of HUVECs growth was inhibited in different degrees after HUVECs were incubated with OX-LDL for 6 hs, 12 hs and 24 hs respectively (P<0.05, as compared with the normal incubation). LF, IP-LP and IP-BC could increase the activity of HUVECs growth (P<0.05). Conclusion LF, IP-LP and IP-BC can increase the activity of the growth of OX-LDL-damaged HUVECs, and the effects of LF tend to be stronger than those of the ingredient pairs.
