摘要 为建立体外检测血小板生成素(thrombopoietin,Tpo)生物活性的生物检定方法,采用MTT法观察了转染Mpl受体基因的TD-3细胞株依赖于Tpo浓度的增殖特性。通过观察实验条件对增殖的影响获得了最佳实验条件,比较了TD-3细胞对10种与巨核-血小板系造血相关的细胞因子的反应,表明该细胞株仅对Tpo和鼠白细胞介素3(mIL-3)有浓度依赖性的增殖反应,对其它细胞因子均无反应,表现出高度的选择性。检测Tpo的ED_(50)为1-3ng/ml,与文献报道检测Tpo生物活性的MO7e细胞株相近。以上特征表明该细胞株可作为检测Tpo生物活性的工具。 Thrombopoietin(Tpo) concentration-dependent proliferation of TD-3 celis of Mpl receptor gene transfected celi line was observed by means of MTT method for the purpose of establishing a new bioassay method to determine the biological activity of Tpo in samples or products in vitro. An optimized condition was obtained after comparison of various experimental conditions. The proliferation of TD-3 celis was highly selective with cy-tokines, only Tpo and mIL-3 promoted proliferation of the cells, but human IL-3 and other 7 megakaryocy-topoiesis related cytokines had no effect at all. The sensitivity of the assay was comparable to MO7e cell reported in the literature, the ED50 for Tpo was 1-3 ng/ml. The results indicated that TD-3 celi line could be used for bioassay of Tpo.
