摘要
利用原生质体融合技术 ,使碱性蛋白酶生产菌 2 70 9与枯草杆菌BD1 0 5(含有碱性蛋白酶基因克隆载体 pDW 2 )杂交 ,得到 1株高产碱性蛋白酶的工程菌A1 6.A1 6的表型与生长特征与 2 70 9相似 ,相同条件下发酵酶活性比 2 70 9菌株高5 0 % ,摇瓶发酵的产酶水平最高可达 30 0 0 0u/mL .用菌落的原位杂交鉴定出该菌株携有双亲的遗传物质 .
A engineering strain with a high ability of producing alkaline protease was obtained by fusing protoplasts of Bacillus licheniformis 2709 and Bacillus subtilis BD 105 [pDW 2(pMK 4 and alkaline protease gene)]. Cell proliferation aspect and phenotype of A16 was similar to that of 2709, and its activities of fermentation were 50%, higher that that of 2709 under the same conditions. The maximum activities of protease were 30000 u/mL from A16 in the optimum fermenntation. In situ hybridization data indicated that A16 carried genes from BD105 (pDW 2) and B. licheniformis 2709.
