摘要
目的探讨不同浓度下紫草提取物对子宫内膜异位症大鼠炎性因子、凋亡基因的影响。方法取SD大鼠40只,均采用腹腔注射苯甲酸雌二醇(0.1 mL/kg)方法制作子宫内膜异位症大鼠模型,将造模成功大鼠随机分为4组,每组10只。紫草提取物高浓度组每天灌胃紫草提取物5.6 g/kg,紫草提取物中浓度组每天灌胃紫草提取物2.8 g/kg,紫草提取物低浓度组每天灌胃紫草提取物1.4 g/kg,对照组则灌胃等量的生理盐水。4周后观察各组大鼠异位病灶组织匀浆中肿瘤坏死因子-α(TNF-α)、前列腺素E_2(PGE_2)、白细胞介素-6(IL-6)及凋亡基因Bcl-2和Bax表达情况。结果与对照组相比,紫草提取物各组大鼠TNF-α、PGE_2、IL-6及Bcl-2表达量均明显降低(P均<0.05),凋亡指数及Bax表达量均明显升高(P均<0.05),且紫草提取物高浓度组TNF-α、PGE_2、IL-6及Bcl-2表达量均明显低于紫草提取物低浓度组、紫草提取物中浓度组(P均<0.05),凋亡指数、Bax表达量均明显高于紫草提取物低浓度组、紫草提取物中浓度组(P均<0.05)。结论紫草提取物可降低子宫内膜异位症模型大鼠异位病灶内炎性因子水平,调节子宫内膜异位细胞凋亡基因表达量,且紫草提取物高浓度作用更好。
Objective It is to investigate the effects of lithospermum arborescens extracts(LAE) under different concentrations on the inflammatory and apoptosis genes in endometriosis rats. Methods Forty SD rats were selected and injected intraperitoneally with estradiol benzoate(0.1 mL/kg) to make rat models of endometriosis. The successful models were randomly divided into 4 groups, each group had 10 rats. The high concentration group of LAE was intragastrically administered 5.6 g/kg per day, the medium concentration group of LAE was intragastrically administered 2.8 g/kg per day, and the low concentration group of LAE was intragastrically administered 1.4 g/kg per day. The control group was given an equal amount of normal saline by gavage. After 4 weeks, the expressions of tumor necrosis factor(TNF-α), prostaglandin 2(PGE2), interleukin-6(IL-6) and apoptosis genes Bcl-2 and Bax were observed in the ectopic lesions of rats in each group. Happening. Results Compared with the control group, the expressions of TNF-α, PGE2, IL-6 and Bcl-2 in every LAE group were significantly decreased(P<0.05), and the apoptotic index and Bax expression were significantly increased(P<0.05), and the expression levels of TNF-α, PGE2, IL-6 and Bcl-2 in the high concentration group of LAE were significantly lower than those in the low and medium concentration groups of LAE, the expressions of apoptotic index and Bax were significantly higher than those in the low and medium concentration groups of LAE(P<0.05). Conclusion LAE can reduce the level of inflammatory factors in ectopic lesions of endometriosis model rats and increase the expression of endometriotic cells, and the high concentration of LAE is better.
