摘要 目的 探讨Dosper和Lipofectamine两种脂质体转染载体的特点 ,为广泛应用打下基础。方法 将新构建载体分别用Dosper和Lipofectamine在有血清和无血清情况下转染包装细胞PA 3 17,通过NIH 3T3细胞检测各组病毒滴度。结果 在无血清组 ,Lipofectamine转染效率略高于Dosper ,但两者无显著性差异 (P >0 .0 5) ;在有血清组 ,Dosper组转染率显著高于Lipofectamine(P <0 .0 1)。两者在有血清组的转染率均低于无血清组。结论 在无血清存在下 ,Dosper和Lipofectamine转染效率都 ObjectiveTo evaluate the transfection characteristic of Dosper and Lipofectamine to estab lish the basis for future massive application . MethodsThe re troviral vector inserted SV40LT antigen gene was transfected into PA317 cell lin es by Dosper or Lipofectamine mediation with or without serum. The viral titer w as determined with the NIH3T3 cells. Results In without -serum condition, the transfection efficiency in Lipofectamine group was higher th an that in Dosper group, but no significant difference(P>0.05); in with ser um condition, the transfection efficiency in Dosper group was significant hi gher than that in Lipofectamine group (P<0.01). And the transfection effic iency in both groups with serum were significant lower than that in both withou t serum groups. ConclusionsTransfection efficiency are high in using Dosper and Lipofectamine in without serum condition.
