摘要
建立了兽用复方磺胺对甲氧嘧啶片含量测定的高效液相色谱法(HPLC)。试验采用C18色谱柱,0.1%磷酸-乙腈(85∶15)为流动相,流速为1.0 mL/min,进样量20μL,测定波长240 nm,柱温40℃。采用外标法定量计算磺胺对甲氧嘧啶和甲氧苄啶的含量。结果表明,磺胺对甲氧嘧啶在30~300μg/mL浓度范围内,其峰面积与浓度呈良好的线性关系(R^2=1),平均回收率为99.3%;甲氧苄啶在6~60μg/mL浓度范围内,其峰面积与浓度呈良好的线性关系(R^2=1),平均回收率为99.0%。HPLC方法有良好的精密度、稳定性,能够用于测定兽用复方磺胺对甲氧嘧啶片各成分的含量,对该片剂的质量控制起到重要作用。
A high performance liquid chromatographic method(HPLC)for the determination of veterinary-used compound sulfamethoxydiazine tablets was established here.A C18 column was selected and 0.1%phosphoric acid-acetonitrile(85∶15)was used as the mobile phase.The flow rate was 1.0 mL/min and the injection volume was set as 20μL.The wavelength was 240 nm and the column temperature was 40℃.The contents of the chemicals were calculated by the external standard method.The results showd that,in the concentration range of 30~300μg/mL,the peak area of sulfamethoxydiazine showed a good linear relationship with the concentration(R^2=1),and the average recovery was 99.3%;and that,in the concentration range of 6-60μg/mL,the peak area of trimethoprim also showed a good linear relationship with the concentration(R^2=1),and the average recovery was 99.0%.The study indicated that The present method is of good precision and stability,and may be used to determine the contents of the components in compound sulfamethoxydiazine tablets,which is significant in effective control of the quality of the tablets.
