摘要
目的探讨回药爱康方(AKF)含药血清对人肺鳞癌SK-MES-1细胞增殖及周期的影响。方法雄性SD大鼠80只,按体重随机分为8组:生理盐水对照组(NS),回药爱康方低、中、高剂量(AKFL、AKFM、AKFH)组,环磷酰胺(CTX)组,AKFL、AKFM、AKFH联合CTX组。分别给予相应AKFL、AKFM、AKFH浓缩水煎剂灌胃,连续5d;CTX组和联合组再按20mg·kg-1给予环磷酰胺于第1、3、5天腹腔注射。NS组给予等量生理盐水。上述各组动物取血制备含药血清,体外培养人肺鳞癌SK-MES-1细胞。应用MTT法检测各组含药血清对SK细胞增殖的影响,流式细胞仪进行细胞周期时相分析。结果 AKF含药血清作用不同时间对SKMES-1细胞生长有抑制作用(P<0.05)。AKF联合CTX含药血清能够抑制SK细胞增殖,其中AKFH联合CTX组的抑制率明显高于其他组,48h之内抑制程度呈时间和浓度依赖性。5%、10%、20%浓度组的含药血清作用SK-MES-1细胞24、48、72h后,与NS比较,除AKFL外,其余各组抑制率均有升高(P<0.05)。流式细胞仪检测结果显示,AKF含药血清及联合CTX含药血清能使SK细胞阻滞于G1期,随着药物浓度的增加,G1期细胞增多、S期细胞减少,和NS组比较,高剂量以及联合组尤为明显(P<0.05)。10%、20%浓度含药血清组与NS组比较,各组的G0/G1期细胞增多、S期细胞逐渐减少(P<0.05);而与CTX组比较,G0/G1期细胞少于CTX组,S期细胞多于CTX组(P<0.05),各联合组G0/G1期细胞多于CTX组,S期细胞少于CTX组(P<0.05)。结论 AKF含药血清可抑制SK细胞增殖,并与CTX有协同作用,其机制可能与其阻滞细胞于G1期有关。
Objective To investigate the effect of medicated serum contained with Hui medicine AiKang recipe on cell cycle of human lung squamous cell carcinoma SK-MES-1 cells. Methods 80 SD rats were randomly divided into eight groups,normal saline( NS) group,low and medium and high dose Hui medicine,cyclophosphamide( CTX),low and medium and high dose Hui medicine AiKang recipe combined with CTX. At the same time rats took Hui medicine AiKang recipe,cyclophosphamide and normal saline for 5 days. Medicated serum of Hui medicine AiKang recipe and cultured cells were prepared. The effect of medicated serum contained with Hui medicine AiKang recipe on growth and proliferation of human lung squamous cell carcinoma SK-MES-1 cells were detected by MTT. The target cells were harvested to analyze the cell cycles by flow cytometry. Results Hui medicine AiKang recipe could inhibit SK-MES-1 cell growth at different times. High dose Hui medicine AiKang recipe could significantly inhibit the proliferation of SK cells. The inhibition rates in AiKang recipe groups was significantly higher than those in other groups. Flow cytometry detection showed that SK cells were blocked at G1 stage after drug treatment. With the AiKang recipe dose increased,cell of G1 stage increased and cell of S stage decreased( P < 0. 05). At 10% and 20% concentration of serum containing group,G0 / G1 phase cells increased in each group while S-phase cells decreased( P < 0. 05). In CTX group,NS group,AKFL group,AKFM group,AKFH group G0 / G1 phase of the cells was significantly less than that of the CTX group( P < 0. 05). Conclusion Hui medicine AiKang recipe can significantly inhibit the proliferation of SK cells and SK cells were blocked at G1 stage after drug treatment. It has a synergistic effect with cyclophosphamide. The antineoplastic mechanisms of Hui medicine AiKang recipe may be due to blocking cell cycle and inhibiting DNA synthesis to restrain SK cells proliferation.
