摘要
在马占相思组织培养初步取得成功的基础上 ,采用二次正交旋转组合设计对马占相思增殖培养基进行优化 ,建立增殖率 (Y)对Ca2 + 浓度 (X1)、6 BA浓度 (X2 )及NAA浓度 (X3) 3个试验因子的正交回归模型 :Y =2 .2 80 - 0 .16 8X1- 0 .2 5 9X2 +0 .185X21- 0 .2 10X22 +0 .16 7X23+0 .32 6X1X2 。从模型推知 ,当Ca2 + 浓度为 0 .5 8倍常规MS培养基浓度 (2 5 5g/L) ,6BA为 0 .76mg/L ,NAA为 0 .16mg/L时 ,增殖率达最大值为 4 .32 ,实验结果与预测值相符。
Based on the primary studies on tissue culture of Acacia mangium,the multiplication medium of Acacia mangium was optimized with quadratic orthogonal rotation combination design. The quadratic orthogonal regression model of multiplication rate(Y)to three factors concentration of Ca 2+ (X 1),6-BA(X 2)and NAA(X 3)was established as y2.280-0.168X 1-0.259X 2+0.185X2 1-0.210X2 2+0.167X2 3+0 326X 1X 2. It was concluded from the model that when the concentration of Ca 2+ (X 1),6-BA(X 2)and NAA(X 3)was 255 mg/L,0.76 mg/L and 0.16 mg/L respectively, the multiplication rate (Y) reach its maximum (4.32) which was consistent with the experiment result.
